Probably my favourite part of mushrooms is the agar work and isolating mycelium… idk why but i could do that everyday for the rest of my life… mycelium are soooo fucking fascinating. I had some pan cyan spores i was isolating and had a isolate that would literally eat trichoderma for breakfast but the fucker wouldnt fruit for shit. Worked on it for months. Cloning is fun too. I kinda miss the tinkering with it but will be to busy to take that up again
We’re you using RTV on dish for spore transfers?

I made my own jars and liquid cultures… for that i used RTV for the syringe plugs that i put into the holes i drilled in the lid. I drilled 2 holes in the lids one for syringe and the other for air exchange… some use cotton but i used a syringe cap that allows gasses but not moisture through and sealed then into the lids with RTV. I can find pics of them if you like

With a hood you will not need RTV or ports.

You can start liquid cultures with water and dextrose (sterilized) and just add the spores in front of the hood. I also drop a magnet stir rod in there and pop the jars on a stir plate from time to time.

Agar plates work really well too, but are more expensive and not as quick to use when doing bulk stuff.

With the dextrose / water in a mason jar thing, you can keep it going indefinitely like sourdough starter.

Yeah with liquid culture you dont even need a flow hood ime. Thats why i started doing that.

i like the agar because you can easily distinguish the different sub species. If cloning though or working with an isolate definitely liquid culture gives you more to work with faster… i always use a stir bar in mine also

Most of the recent talk in about Mushroom growing and not tissue culture. Tissue cultures needs similar sterilization but also requires additional prep for the tissue being cultured. It is probably the ultimate in regeneration of an old and tired mother plant

Absolutely H2O hombre. H2 hombre? H2Ombre?

The difference is doing it in bulk and doing it for personal use. For experimentation, plates are the way to go. When you are doing hundreds of pounds - that stuff just slows you down.

For my liquid i used a light malt agar really diluted so the viscosity kept things more suspended with dextrose. But it depends on what ya doing i guess. I also used an extremely tiny amount of turkey trot (turkey poop) boiled into the water then i poured off and pasteurized before cooling and using. That really gave a boost to the colonization rates and the mycelium seems to go crazy with it

I think they loving the o2… those babies are nicest roots i have seen. As the golden arches would say…. da da da da da…. I’m lovin it.

Hands down the most rewarding part of it. Even in a still air box.

You guys really aren't helping extinguish this fire that's about to burn up my pockets with all this discussion.

Looks like my winter got way fuller.

@Moe.Red have you looked into trying cordyceps at all? Super interested in that and you grow it on dead bugs... so my inner child is just like 'uhyup.'
