Here is the way I use brix. (I'm new to this too, love to hear other ideas)
Find a fan leaf that is in direct sun that is sorta average for the overall plant. Not the highest, not the lowest.
Snip it including the petiole and smash it to get the sap out. I use a $2 garlic press. ($47 for anyone who carves pipes.)
Put a drop of sap on the meter, close the lid, make sure the whole glass gets wet when you close it. Does not take much, but if you don't get enough on there wipe off and try again.
Look thru it. You will see a line - basically 2 colors. Where the color changes, read the number, that's your brix. If you have ever used a refractometer to measure salinity in a saltwater aquarium - its the same kinda thing.
Interpretation:
The higher the better. Try for double digits.
Lets say you are at 6. You could try something, wait a couple days, remeasure, and make an interpretation as to the effect your change had on the ability for the plant to make sugar.
Example - Brix is at 6. Lower your lights 6". Brix goes to 7 - good job. Brix goes to 5 - too much light. Can do the same thing with say Nitrogen, PH, foliar sprays, whatever.
When it comes to photosynthesis, the plant will go full bore until it meets a limitation. Could be light, CO2, well, you know how to grow I don't need to spell it all out.
You can use this data to understand what your limiting factor(s) are.
You can also use it to detect your light penetration. Use it to determine if lower vegetation is helping or not. If your top leaves in the sun are double digits, and the lower leaves are 2, they are not really helping you. If they are blocking air movement, causing your RH to go up, whatever, you can cut them off knowing they weren't doing anything good anyhow.
I'm gonna go out on a limb and say lots of the growers on this thread can intuit what is going on with the plant without needing hard data. Definately not necessary for everyone. But if you are in there trimming fan leaves anyhow, why not do a gut check as to what is going on with the plant? It's only $19 unless you live in siberia, in which case screw you in particular, gimme $36.
